Zielgruppe: Studierende der Chemie im Masterstudiengang;
Art der Durchführung: Vorlesung mit Exkursion zu Laboratorien der BAM; die Veranstaltung schließt mit einer Klausur oder einer mündlichen Prüfung ab.

Gegenstand: Es werden gängige material- und bioanalytisch relevante physiko-chemische Verfahren vorgestellt mit dem Fokus Qualitätssicherung und optische Spektroskopie. Dabei werden die Grundlagen zur Qualitätssicherung mit Methoden der physi-kalischen Chemie an Beispielen behandelt wie der Charakterisierung von optischen Messgeräten inklusive radiometrischer Rückführung, Anforderungen an Standards, Aufstellung von Messunsicherheitsbilanzen und relevante Normen. Am Beispiel der Fluorometrie wird der Einfluss von probenspezifischen Effekten auf signalrelevante Messgrößen für verschiedene Chromophortypen (molekulare Systeme, Nanokristalle, Partikel) dargestellt und Auswahlkriterien für Chromophore abgeleitet.

Deliverables: Lecture with excursion to laboratories of BAM; the lecture includes either a written or an oral examination.
Prerequisites: None required;
Content: Physicochemical methods commonly used for material analysis and bioanalysis will be presented with the focus on quality assurance and optical spectroscopy. The basic principles of quality assurance with physicochemical methods will be discussed for examples like the characterization of optical measurement systems including traceabiliy to radiometric scales, requirements on standards, uncertainty budgets, and relevant written standards. Exemplary for fluorometry, the influence of sample-specific effects on signal-relevant quantities will be discussed for the different types of chromophores (molecular systems, nanocrystals, particles) and criteria for the choice of suitable chromophores will be derived.
